Thanks, Kpmac. The micrometer-manipulated stage helps greatly reduce the frustrating part of the process. This is important because as my age goes up, my tolerance for things that annoy me goes down.

Curmudgeon, thanks for asking. The specimen remains stationary once set in place. The camera is mounted on a digital motorized focus rail that controls the start and stop distance that the camera will travel and the increments between each shot to be taken based on the depth of field of the optic or lens used.

This session is programmed to travel three microns, stop, wait three seconds, take a shot, wait to seconds, and move to the next step of advancing the camera forward. The three-second waiting period is to let any vibrations from moving cease before releasing the mirrorless shutter. The two-second waiting period after activating the shutter is to allow time for the buffer to empty for the shutter speeds can be a second or two depending on the attributes of the specimen (bright colored, reflective, dark, hairy, etc.).

Once everything is set, it becomes a "lights out" operation with the only light in the room focused on the specimen. That's how the black backgrounds and transient reflections of unwanted light are accomplished.
