Corkscrew Swamp Sanctuary would be at the top of my list followed by Ding Darling park, Barefoot Beach and of course Everglades National Park.
I recent spent most a week in the Ft Myers area. Depending on how much you want to travel, consider:
a) the burrowing owls around the Cape Coral public library.
b) sunset at the Ft Myers Pier
c) Sanibel Island Lighthouse
d) JN Ding Wildlife Refuge
e) Corkscrew Swamp Sanctuary - the ticket is 2-day, so don't feel rushed for all in one day.
f) Naples Botanical Garden
g) Ringling Museums (Art, Circus) - Sarasota
Good lists here. Winter brings a lot of birds. Ding Darling is great as is much of Sanibel Island. The nature reserves are also great.
The 6 Mile Cypress is nice too -

The Edison and Ford Winter Estates for starter (see photos below).
Have a drink or 2 at Nervous Nellie in downtown Fort Myers Beach while enjoying the gulf scenery (see photo). Enjoy the downtown walk and the souvenir stores on both sides. Parking may be a problem and road construction, may be, as well.
Check the neighboring areas, too, for great shooting opportunities: To name a few, Sanibel Is. (be ready for long lines at the toll bridge), Marco Is. (Snook Inn for dining, drinks, and nice view of high-end boats but busy place), Punta Gorda (nice ride to enjoy well-maintained, quiet community with high-end houses), and Naples (watch out for a bunch of Bentleys prowling). If you got extra time to unwind, head to the Naples Pier and enjoy the scenery with option for people-watching in different sizes and proportions.

Anyway, enjoy your vacation. (Temps in the 80s. I'm jealous!)

Lovers Key State Park. An abundance of wildlife, especially birds. Wading birds and Ospreys are very common and approachable.

You’re not that far away from the Ding Darling nature preserve which is well worth your time. The Edison and Ford homes.
I just went out to Babcock Ranch and got some nice shots on the wildlife eco tour.
Nesting bald eagles up on Bayshore Avenue...google eaglecam North Fort Myers for details. About 40 yards for the road.
Sunsets at Blind Pass (between Sanibel and Captive)......
For us, it's a toss up between the Atlantic Coast and the Gulf Coast for Florida, but we have done FM, Marco Island and Sanibel. If you like birds, this is a superb location, nad the suggestions above largely focus on birding. We've done most of these,a nd I;d add a lng distance UK 'tick;' as well.
One I would add is Marco Island/Tigertail beach - easy burrowing owl sighting, lots of other birds. Have a look at this page on our web site.
