You probably have an option for an airboat tour of the everglades. You may not have a lot of wildlife photo ops depending on time of day. Best to leave the 600 and try a 70-200. Not much time to set up a tripod!

Just visited the Dry Tortuga National Park in December. You would be able to see birds for sure at this location. But its in the middle of the Gulf of Mexico and its a full day excursion, leaves at 8 in the morning (or thereabouts) and doesn't return until 5sh. Its an interesting place and beautiful if you have the time. Other than that Key West is a great little town but I wouldn't want to lug that 600mm around. Check out Mallory Square at sunset. Bon Voyage!

At Key West see Emerson House for sure. Bring a 33mm lens or wider. Bars: Hag Breath, Irish Pub, Sloppy Joes. Garden of Eden.

Key West has very colorful architecture and there are wild chickens and roosters all over the place, many of which are colorful and photogenic. Not sure you would need your 600mm prime lens here, unless you climb to the top of the lighthouse on Whitfield Street near the Hemingway House, which affords some great photo opportunities.

Depending on how long you have in Port Canaveral, it’s a relatively short drive (if you have access to a car) to the Merrit Island Wildlife Refuge (Eagles and gators). Kennedy Space Center is a popular tourist destination, but a little-known spot is the space museum on Cape Canaveral Air Force Station - go to the South Gate and request a pass. There’s a huge assortment of interesting rockets, a 60s-era blockhouse for launches, completely equipped and a museum.

In Charleston, consider the historic downtown area along the Battery, Patriot’s Point (the Carrier Yorktown, plus the Destroyer Laffey and a WW2 submarine) and Middleton Plantation (again, a car will be required)

Street shooting is fantastic there. Also take pictures Of the boats and yachts. And fish that people on the boats catch.

If you have time in Port Everglades you are about 60~70 miles (~100 km) from Everglades National Park, one of the most amazing places anywhere! At the north boundary of the park is the Shark Valley Visitors Center. There are short tram tours with guides who point out wildlife and talk about the park. There is an observation tower on the tour and alligators are practically guaranteed!
If you go South into the park you quickly reach Anhinga Trail, a short hike that almost always has tons of birds and alligators. Just Don't get too close to the 'gators, they are blindingly fast if you annoy them!
Your 600-mm and a tripod will be great for Anhinga Trail and your gear will not even particularly stand out in this birders paradise!
If you can't do the park you might see if you can arrange a swamp buggy (generally suitable for photography) or an airboat ride (exhilarating! Leave the 600-mm on shore!) at Billie Safari operated by the Seminole native-American tribe.

At Port Canaveral you will not be too far from the Merritt Island National Wildlife Refuge. But even more important you will be right near the Kennedy Space Center! Totally worth a visit! Wide-to-normal to short telephoto lenses will probably serve you best at KSC, longer lenses for Merritt Island NWR.

I would leave the 600mm at home and bring something else. Why lug that on a trip and be a target?

Of the places you mentioned, I've only been to New Orleans and Charleston. I went to the 1984 Louisiana World Exposition in N.O. and stayed in a hotel in the French Quarter. It was an interesting time. (Unfortunately, I don't seem to have digitized my photos of that visit.) Charleston, SC has many things to see, including the plantation that was used in the movie Gone With The Wind--check out the various excursions available.

When we were in Key West, we took a tour on a small boat that said we would probably see dolphins, and we did. We were alongside a group of them for several minutes and got some good photos. Sorry I don't remember the name of the boat, but a concierge in one of the hotels should be able to give you some suggestions.

Charleston is great but I wouldn't go there for the wild life. Bring a wide angle and capture the historic district. Lots of churches, restaurants, and some great old houses.
