I just bought a Opteka 420-800mm lens. What is the best amp and shutter speed for day and night use.

There is only a best shutter speed and aperture setting for the situation on hand ( does not matter, if it's day or night, it all depends on "what" you are shooting and how you want it to look)!

Since it's an HD lens you don't need to set the aperture at all. It set itself depending on the zoom setting. Set your camera on A (Av if you have Canon) and the shutter speed is set automatically.
Can't say what kind of IQ the $150 super telephoto zoom would deliver.

That's an F/8.3, so you're going to need a lot of light. Owners seem to love it.

I am sure he means aperture but it's hard to say for sure. Maybe he is a closet electrician and can't get amp out of his mind. Using the abbreviation, amp, for aperture is shocking to say the least.

15 amps should be enough
