Is it necessary to use the same card in the same camera at all times? Have a canon t2i and nikon d600 seem they dont mix well.. any thoughts?
You would need to format the card for each camera. Once the card is formatted for the Canon it won't be able to be seen by the Nikon.

You can interchange them all you want, each camera will form a folder for its images.
I have cards I use in my sales cameras and many of them have folders for 6 or 8 different cameras on them with no problems at all, Nikon, Canon, Sony and even Pentax folders all reside on the card separate from each other. One thing I have also noticed is that a Nikon camera will even display on its LCD screen JPG images from some Canon and Pentax models.

Good advice, as usual, MT! As a computer guy, I might add that it never hurts, once you have several backups, to format the card in the camera it will be used in. You get a fresh file system that way and if there are any problems with the card, better to have them show up during a format than AFTER you've taken a lot of pictures.

I did an experiment and used the same card in several different brands and models. No problem. Each camera just made its own folder for pictures. It's probably not the best practice, but I used the card in different Nikon DSLRs, P&S Canon and P&S Sony.
At the risk of starting a long thread.....
SD cards by their nature are prone to being mishandled (removed while being written for example). This typically does not happen with hard drives. so SD cards are much more likely to have corrupted FAT tables than hard drives.
How to ensure that the FAT table is as pristine as possible? format and then ensure not chance of mishandling....so format in the camera (you still run the risk of having the battery go dead in the middle of the write, but lets ignore that case for now).
Other than the chance of mishandling, I will go on a limb and say there is no difference if you format in camera or out of camera. Both will give you good FAT table formats; I am confident that windows and all the camera makers have that all sorted out. And if the camera does not see the files or directories it needs to write files, it will create them on the fly.
My T1i and T4i do not trade cards, even formatted in camera, they like their own, 2 cards for T1i and 3 for T4i, don't know why, but that's the way it is, Bob.

They should change, both are SD storage.
