I am using post- processing software Lightroom 4, my question is how I save my picture after I color spot one area in a black and white photo in the develop module. I will appreciate any help.

I'm sure you will get better answers than mine, but here goes. In LR, the adjustments you make are not really to the originals. As I understand it, LR writes a file that defines the changes you have added. When you EXPORT the picture, then it writes the changes on the JPG, but never touches the original.

Therefore, after making the picture what you want in develop, export and you will have the jpg with the changes. You don't have to "save" anything while making changes in develop. I guess that is done automatically since if you close LR and open it again, you see what you were working on with the develop settings the way you left it.

Hi Charles and welcome. As another post put it, Lightroom does not do a adjustment to the photo. It creates a sidecar to the image with the adjustments in the sidecar and the regular image. (LR5 not sure about 4). What you need to do is make all of your adjustments then export the image to either a .jpg or a .tiff. This will give you the complete "corrected" image. It will also leave your original so that you can make further edits or restore it to original. You can also make a "virtual copy" of the image before (or at any time during) your edits and make changes to it. This is handy if you know that you will need 2 or more different crops or one B&W and 1 color or whatever of the image (you can make as many virtual images as you want and the do not take up drive space.
