Like a lot of things, it depends. Let us assume that neither partner has major genetic defects, and that the genetic defects they do have (as we all do) are not incompatible in the sense that their immediate offspring are likely to be disabled. This would be the case if you selected any two people of breeding age at random.
In this scenario, there would be more problems with genetic defects (the problem with inbreeding) than is the case in the general population today, at least for the first ten or so generations. But these problems would not show in a majority of their descendants (depending on assumptions made about how many children each generation produces, the rate of genetic change, etc.)
Thus it would be possible for one couple to repopulate the earth. The greatest risk would be in the first few generations, when a small population would be most vulnerable to natural disaster, famine, disease, predation, and other risks.
