I have had a 5dsr since it went on sale. I set it up the same way I set up my 1ds iii. I only use back button focusing, I turn off focusing with the shutter button, I only focus with the center sensor on the spot I want in focus, and then move the image to the scene I want. I generally use the matrix meter, although I use spot metering for some lighting situations. I usually shoot a low 100 - 200 ISO. The one change I have made to my shooting with the 5dsr is that I generally shoot at higher shutter speeds. The 50 mg pixels really require a steady hand I so not use a tripod a a lot. When I do shoot with a tripod I use a cable release.
I love the camera. The image are great and they allow you to crop in ways that other cameras do not.

CreativeLive is a great place to learn. John Greengo is one of many excellent instructors.

For anyone interested, they are having their "Photo Week" starting October 9 through October 13. Look for it in their "On Air" module, and you can access the information about it. They publish the schedule for classes for each day so viewers can choose what they want to see. If you want to sign up, you can RSVP. It is free. I know the information page says it costs $200, but that is for the video of ALL classes, a sale price only available during the week, then it goes up.

Hard to give any advice without knowing more about the type of photography that interests you. The reason there are settings is to make it possible to adjust the camera to better fit a particular purpose. Without knowing the purpose, it is impossible to recommend settings.
