Wheel lock sounds like what is found on an ISO standard hot shoe.

Minolta used those until the late 1980's when they introduced the Minolta mount which did not use a wheel lock.
My flash has a wheel that that acts like a lever that pushes and releases a pin to lock the flash

Now, some 3rd party manufacturers DID use a cheap wheel lock instead of the Minolta auto-lock. I have a Poloroid Ring Flash (made in China) for a Minolta hot shoe which has a thumb wheel. It's a piece of crap.

Just to add some more confusion/data to the situation:

I've been attempting to do this in S mode with the shutter set to 1/125.
When I hit the "test fire" button on the transmitter, the light on the receiver comes on (and off, just as one would expect).
Also, I have a SIGMA EF-530 DG Super which is behaving the same way.

This may seem an oddball question... but which firmware version do you have installed on your camera?
