In Central Florida (Lakeland) you MUST go to Circle Bar B Preserve! VERY accessible for non-bushwhackers(My brother-in-law rides his bike around the trails). Birds galore, 'gators, armadillo, even wild pigs. (Oh, and the raccoons!

We rent a place in Everglades City and take The Loop. A great way to optimize your time/wildlife ratio. We usually go in March.
Also, a flat-bottom boat tour on the Glades from Everglades City ($150/person) sounds steep but I got great close-up shots of birds in their habitat. Well, close-up with a 70-400mm lens. Do not go to Florida without a good tele-zoom!
The wife loved it. With the boat tour, we stopped and did some shelling on a remote island (had its own porta-potty). In both the Loop and boat, there is so much wildlife that your wife will be entertained. On the Loop, everything can be seen from the road, no hiking involved.
The national park entrance at Homestead is also something your wife would enjoy, nice paved walking trails and boardwalks in the swamp with lots of birds and gators. Permanent restrooms there.

Sanibel. Great vacation spot, lots of amenities. But also has Ding Darling National Wildlife Refuge. Outstanding nature photo opportunities abound. In addition, another nature preserve, the Corkscrew Swamp Sanctuary, is an easy drive from Sanibel. Don't be put off by the "swamp" part - it's mostly boardwalk.
Best times - late winter or spring.
I too have spent countless hours in both places, another good spot would be Lovers Key State Park, also close, lots of wildlife, and a good place for the wife to stretch out ans sun whilst you take photographs.
