Having lived in that area for eight years, I believe the best are amongst those mentioned above. Watkins Glen, Taughannock Fall SP, The entire Finger Lakes area - particularly the wine country around Hammondsport (not just about the wineries - the lakes and hilly, scenic countryside are beautiful! It’s hard to go wrong. I’m not much for museums, but the Glenn Curtis museum on a rainy day might be a great break. The area is full of excellent restaurants too.
Distances are small enough to plan around your interests, with only moderate attention to geography. Find a centrally located motel to stay at, to avoid continuous packing/unpacking. I like Skaneateles at the north end of the lake of the same name, but it doesn’t qualify as central.

Magnolia Place in Hector is a nice B&B. You might also enjoy the Rockwell Museum in Corning.

My wife are planning on a fall trip to the Finger Lakes and Adirondack. We are planning around first couple of weeks of October.
We are thinking of Watkins Glen, Letchworth St park. Suggestions on other must stops and maybe some helpful suggestions on lodging. We are not campers, being in our mid 70s. thanks

I will add Robert H. Treman State Park, 105 Enfield Falls Rd., Ithaca, NY. I did a trip to this area a few years ago. This park was well worth a visit.

If you pass through the Syracuse NY area, visit the historic village of Skaneateles. If you are Kayaker's, Hemlock lake is pristine, no cottages permitted anywhere near the lake, and boat motors limited to 10 horsepower max.

For a good view of the surrounding mountains and foliage, Coney Mountain just south of Tupper Lake on Rt. 30 is an easy climb of about 1/2 hour. Bog River Falls is nearby and looks especially beautiful in the fall.

The Cornell Universit6 campus has some beautiful spots. Bridges over the Gorges are pretty neat. Beebe Lake, Sapsucker Woods.

In the Adirondaks Lake George is a must, IMHO, Ausable Chasm, on my list we took the kids there long time ago, Check out "Only in New York" site sends you interesting things to do in NYS, not a state gov site.

In the Adirondacks there was a museum at Blue Mountain Lake. If it is still there it is probably worth going to.
