I have a Canon 80D. I have the option to use a Canon macro EF 100mm 1:2.8 USM or a Tamron 16-300mm 3.5-6.3 Di II PZD macro. Which lens would be better to use and would extension tubes help? Would a 50mm lens be better?

How close do you need to get? Most of the time when someone without macro experience thinks they need a macro lens what they really need is a lens that allows closeups, maybe up to 1:3 or slightly higher magnification. You would need a macro if you wanted a full-size image of the stigma or the anthers. If you want to show a whole flower, you don't need a macro lens.
