My largest backpack is a LowePro Photo Trekker. It can definitedly accommodate a 300/2.8 with camera mounted (or up to 500/4 unmounted), with hood reversed. It also has room alongside that can handle a couple more camera bodies and lenses, but not with the lenses mounted.
Frankly, the Photo Trekker (an older model.... I think it's most similar in size to their current Pro Trekker 450 model:
http://www.lowepro.com/trekker) is the largest and heaviest backpack I'd ever try to lug around. I have hiked with an unmounted 500/4 in it, along with other lenses, 2nd camera, and a tripod in a separate shoulder bag. The backpack with this gear weighs over 30 lbs (the 500/4 alone is about 8 lbs, tripod with head & accessories weigh an additional 9 lbs). About 40 lbs total! It is a definite chore to carry it very far!
I much more often use a smaller Mini Trekker. That can accommodate up to a 300/2.8 unmounted and hood reversed, plus a couple other moderate sized lenses, or a second body and a smaller lens or two.
I usually have another camera and lens ready to shoot, outside the backpack. Plus I often have a vest on, to corral some of the smaller stuff such as spare batteries and memory cards.
When travelling by air, I use the Mini Trekker because I can carry it on and it will fit into an overhead bin (the Photo Trekker is too big). When loaded with a 5DII w/battery grip, 300/4L IS, 24-70/2.8L, 20/2.8, 135/2, flash, light meter, a few 72mm and 77mm filters, extra batteries, memory cards and other misc. accessories, it is fully stuffed and weighs close to 25 lbs, which is about as much as I'm willing to hike through airports carrying while also wrestling one or two checked bags with clothes and such.
