I've had several cameras with U1 and U2 settings, but I've never used them. How do you people use them?

Jerry, I have U1 programed to use off camera flash. This gives me a quick way to get to it without having to go into the menu and configure everything on the fly.
I have U2 programed for Timed exposures. Again it keeps me from stumbling around in the dark trying to configure the camera.
My camera strap has pouches for extra batteries sd cards etc so I keep a note card that reminds me how I have U1 and U2 set. (I always carry a pocket flashlight and have a spare in the camera bag.

I have one of them set up for my usual macro settings so even when I change them during use I know they will return to my settings when I turn off. The other one I use sometimes for whatever I am playing with at the time.

Good idea.

I use my D7200 exclusively with my 200-500mm and I have U1 programmed for birds in flight and U2 programmed for wildlife. This way as I'm out and about and I see one or the other I can quickly switch to either/or from manual (where I normally shoot) so as not to miss quickly disappearing opportunities. I often adjust the parameters from the default settings on the fly if necessary but in a pinch it's ready to shoot.

The Canon sx50 may not have the choices I'm reading here but I use the shortcut button to toggle among jpg, raw, jpg+raw.

My new-to-me Lumix DMC-G7 has four shortcut buttons. So far, I haven't been able to remember which ones I set for what, and sometimes have to hit more than one to find my goal quick ISO adjustments, exposure compensation, stopped-down dof check, and something else

I have one set up for macro, and the other for non-CPU lenses, I think. I never use them.

Both are set for variations of Manual with back button focus. U1, I use most often, especially for any form of flash photography. U2, has a different metering mode and uses auto-iso (for non-flash).

AF is set to the default 'release button' in all other modes so the wife, who doesn't do BBF, can also use the camera.

They are very useful. With my Nikons I set up some variations in the settings I use for studio shots. A typical studio shot for me would be in manual mode with ISO 100, 1/125 second shutter speed, and f/8 aperture.

So you can toggle BBF on and off? Clever.
