Hi, like to get sharper focus anytime. But to do the adjustments do the say dollar bill have to be up to or over 40 feet away from camera?

No but that is the minimum distance when adjusting a 500mm lens.

To help you out look up Lens Align and watch some videos on YouTube. Lens Align will give you the minimum and ideal distance for setting your target. One other thing to look up is the "Green Dot" focusing system.
